POST-İSKEMiK BEYİN HARABİYETİNİN AZALTILMASINDA ALLOPURİNOL'ÜN ETKİNLİGİ

Geçici ön beyin iskemisi oluşturulan sıçanlarda ksantin oksidaz inhibitörü olan allopurinol'ün nöron harabiyetinin azaltılmasındaki etkinliği araştırıldı. Erkek Sprague-Dawley sıçanlara karotid arter oklüzyonundan 48, 24 ve 1 saat önce allopurinol (SO rng/kg) uygulandı. Bir saatlik sol karotid arter oklüzyonunundan 24 saat sonra hayvan feda edildi. Beyinleri hızlı bir şekilde çıkartıldı ve serebral ödem miktarı, infarkt hacmi, lipid peroksidayonunun bir göstergesi olarak MDA düzeyi, süperoksit dismütaz (500) ve Katalaz aktiviteleri belirlendi. EFFECT OF ALLOPURINOL IN DECREASING POST ISCHEMIC CEREBRAL DAMAGE

The effectiveness of allopurinol which is a xanthine oxidase inhibitor, for decreasing neuron damage of rats which temporary precerebral Ischemie had been constituted, has been investigated. 48, 24 and 1 hour before carotid arter occlusion, male Sprague-Oawley rats have been administered allopurinol (50 mg/kg). After one hour, rats have been sacrified, after left carotid occlusion. Their brains have been removed very quickly and the quantity of cerebral oedema, the volume of infarct, MDA level as the indicator of lipid peroxidation, superoxide dismutase (500) and catalase activities have been detennined. The quantity of post ischemic oedema of the rats to which allopurinol was applied has been deueased significantly, compared with that of the rats under control (p<0.001). Important decreases (p<0.001) in infarct volume and lipid peroxidation have accompanied to the decrease in cerebral oedema, and im portant correlations have been found. Reverse relations have been found between SOO, catalase activities and infarct volume. Post ischemic SOO and catalase activities in rats to which allopurinol was applied have been found significantly high (p<0.001), compared with that of rats urıder control. These fındings indicate that allopurinol has effect in decreasing the post ischeınic cerebral demage.
